耐熱性酵素生產菌Bacillus Subtilis WLA12 之分離與定性

本實驗針對一取自台北縣烏來南勢溪下游之溫泉菌,進行微生物學、生物化學、分子生物學三方面之觀察與實驗,期能得到一可生產特定酵素之菌種,且具耐高溫之抗逆境能力。對其酵素進行定性,並嘗試轉殖出相關基因,使之可大量表現。目前已篩選出一種可生產多種酵素之菌種〔依其採集地點暫稱WL-A12〕經菌種鑑定為Bacillus subtilis WL-A12。藉由菌落檢測法以及Zymogram 的方式做酵素分析,並以電導轉形等技術,希望能成功轉至E. coli 上表現。另外,也對該菌種作了一些基本微生物方面的觀察〔如需氧情況、最適生長溫度〕。We isolated enzyme-producing thermophilic bacteria from hot springs near downriver of Nan-shi, Wulai, Taipei (北縣烏來南勢溪). Through microbiological, biochemistry and molecular biological analyses, a multiple enzyme-producing Bacillus subtilis strain, designated WLA12, has been isolated. The growth condition of WLA12 was observed. Using basic colony assay and zymogram analysis (gel electrophoresis) to observe the expressed enzymes, molecular weight and gene size of the enzymes were revealed. With comparison to E. coli control strain, the related enzymes were only found in WLA12. To express the Bacillus genes in E. coli, molecular cloning and gene transformation via electroporation was carried out.

臉上真的有蟲嗎?~揭開蠕型?的真面目

I’ve heard a report on the internet that there are mites on the face, even a clean face, and I took an interest in doing a research. First, I found that on the face is Demodex, which lives and depends on human beings; besides, nearly seventy percent of people have these mites on their faces. To know more about the habit of Demodex, I sampled forty people of both sexes and different ages. The analysis, not correlating with sex and times of face washing, showed that more mites are found on the forehead than on other parts of the face, and there is more probability to find Demodex on the face of those who are older, those who have oily skin, and those who suffer from acne. I also observed that these mites are photonegative, often gather together in the hair follicles, and feed on sebum. Moved from the human body, Demodex is livelier in sesame oil than in other kinds of oils, but its life ends in about eight to ten hours and dissolutes at last. In addition, I tried to devitalize Demodex by various kinds of medicine, cleansers and cosmetics, but only those containing sulfer, eau de parfum, and the essence of rosemary or lavender are efficacious. 網路傳聞乾淨的臉上也有蟲,引起我探索的興趣,查探後發現是和人類片利共生的毛囊 蠕形?和皮脂蠕形?,初步調查顯示近70%的人臉上都有蟲。為了更了解蟲的習性,以年齡 與性別區分共在40 人臉上採集樣本,統整結果得知,額頭比其他臉上部位多、年齡越大、膚 質油的發現機率較大,但不受面皰多寡、性別、洗臉次數等影響;觀察後發現蠕形?以人類 的皮脂為食,經常群聚在人體毛囊中,離開人體後在芝麻香油中活動力較佳,但約8~10 小時 後就死亡分解導致無法繼續觀察其生活史,對光有明顯的負趨光性,最後試著用各種藥品減 低蟲的生命力,以薰衣草、迷迭香精油、毒藥香水和蜜花沉澱硫等較有效。

平面上三點集中度判別法之探討

關於平面上若干點的集中度之定義,一直很少有人予以仔細的探討,因為判別的方法有很多種。本研究是以平面上三點所構成的三角形之三頂點作討論,分別以三角形的面積、周長,內心、外心和重心至三頂點距離,三點的標準差及平均差的概念,作為不同的判別方式,並以動態幾何化版GSP模擬不同的判斷方法做為研究,在利用統計學上的方法,找出哪一種定義方式最為適合、誤差最少的判斷方法。There are many ways to determine the centralness of three dots on a plane; however, no definition study has been applied. In this study, we focus our interests on the entralness of three vertices of a triangle formed from three dots on a plane. Various methods such as the relationship of area, and the distance of the triangle to the centralness of three dots, the distance from the interior point, exterior point, the barycenter to the vertices of triangle, and the standard error and differences average obtained from three dots were determined with a dynamic geometry software GSP, and a statistic method was used to find a least error way to determine the centralness of three dots on a plane.

應用吸水高分子螯合重金屬離子及奈米銀的製備

聚丙烯酸是尿布中吸水成份,若被隨地丟棄勢必對環境造成污染。分析其結構,由於存在對重金屬離子具螯合作用的羧基,因此,可用於捕捉重金屬離子,作為偵測污水離子的利器。另外,若將螯合的銀離子以化學還原法製成奈米銀,將可應用於抗菌。研究結果有: \r (1)PAA對Mn、Fe、Co、Ni、Cu、Zn、Ag等離子具有明顯螯合效果。 \r (2)螯合反應:PAA+Mn+ [PAA-M]n-x+xH+,其可利用濃鹽酸使其再生。 \r (3)螯合能力以Fe3+最強,Zn2+(Mn2+)較弱。 \r (4)螯合離子的最小極限為10-3(M),我們採樣的廢水有重金屬污染,推測濃度大於10-3(M)。 \r (5)成功將螯合的Ag+製成奈米銀,為金黃色,屬於球形銀,甲醛濃度愈高,奈米粒徑愈小。 \r (6)奈米銀與銀離子在抗菌上的確有明顯效果,帄均粒徑小,抗菌效果越強。

聽音辨位--聲波的測量應用

本實驗設計主要是以波的傳送速度(特別是聲波),以及接收收到的時間值來做實驗、運算、討論。而其特點是為了應用於實際生活中,做了許多異於平常測量方法的設計。主要是使用時間差(|t1–t2|V=發聲器到兩感應器的距離差 )來消彌掉一般測量時,需要採取同步的條件,說明如下: 1. 由以上的圖中,t1’ = T + t1 為實際由感應器開始感應到感應器#1 接收到訊號的時間;同理,t2’ = T + t2 為實際由開始感應到感應器#2 接收到訊號的時間。而T 為感應器開始感應到發聲器開始發聲的的時間(之後的 T 皆為如此)。由以下式子得知: |t1’ - t2’|=|( T + t1 ) - ( T + t2 ) |=|t1 - t2|及為本實驗所需的時間差。利用減法將T 消除,便及為發聲器與感應器不必採取同步,此為本實驗目標以及優點之一。 2. 之後的公式推導中,實際由感應器開始感應到感應器接收到訊號的時間中,表示為t1、t2、t3……以此類推。 像是市面上販售的反射式測距器由於其直線性的限制,在我們可負擔的情況下,就只能做一維的測量,而在本實驗中,我們使用多個感應器,而可測量至二、三維空間,並使測出的物體由相對位置轉為絕對位置。再加上正在計劃中的測量儀器改良與自製,例如利用電腦的音效卡接上麥克風或是其他感測器,以及電子零件、電路的組合與設計。而在於一般的實際應用面上可配合工業的破壞性檢測,甚至是橋樑的斷裂處、各種振源的測量,亦或是人員的搶救,都應有不錯的效果與利用價值。 1.The major design of experiment is to spot the location of an object by experiment, calculating and discussing of such figures like the transmission speed of the waver (especially sound wave), plus time value of the receptor and so on to get the result. 2.In practice, the ordinary measuring method has to be implemented under the circumstance of synchrony: however, the distinguishing characteristic in the experiment is to overcome such restriction with the use of the “time lapse” concept. 3.The reflecting measuring instrument on the market is limited by its “straight-line characteristic.” Instead, we use multiple sensors to spot the absolute location of an object in its 1-D, 2-D, 3-D form. 4.We have now been working on the improvements of the measuring instruments, for instance, using sound cards to connect to the microphone to make a new sensor; also, the redesign and combination of other electric parts and circuits are also under construction. 5. We plan to apply the experiment not only in spotting the location of an object but also in further spotting the location of vibration coming from various objects (e.g. in the use of rescue).

橘子甜了

從研究抑制乙烯的實驗中碰巧得到的靈感,讓我們找到了水果中一種不可思議的變化,水果在撞擊之後乙烯量會增加,因而帶動水果的糖度上升,甜度增加!! 我們利用水果內的逆境機制,使得水果在外界刺激之下(如:撞擊),出現加速成熟的效果。我們經由多次的實驗,在各種水果的數據中,分析變甜的原因,及與乙烯量增加、pH 值下降的相互關係。並且找出除了搖動外,其他可以使水果糖度增加的方式。有了這些方法,我們可以在家中自行加工水果,使未成熟的水果快速成熟、使已經成熟的水果更甜,再也不會因非產季而妨礙到吃的興致!! We get an inspiration form the experiment for controlling ethylene. We find an unimaginably different change of fruits. After ramming, the amount of ethylene in the fruit will increase. This makes the sweet degree of the fruit increase, and it tastes more sweetly!! With adversity system of fruit, we make fruit ripe quickly by external excitement.(ex: ram)Through many experiments and the data of all kinds of fruits, we can assay the reason for fruits’ becoming sweeter, and interrelation between increasing ethylene and decreasing pH value. And find other ways except for shaking to make sugar degree rise. With these ways, we can process fruits by ourselves at home. We can make unripe fruit mature quickly, make ripe fruit sweeter, and we will no longer be obstructed to eat fruit even if it won’t be produced in that season.

國民身分證相片規格驗證暨浮水印防偽系統

政府全面換發國民身分證,並訂定新式身分證之規格,以防範遭不法偽造之情事,確保民眾權益。然而其中的照片規格,有十多條規格的限定,若用傳統的辨別方式,近1876 萬張照片是否合乎規定,那將耗費多少的人力呢?於是本次研究主題「新式國民身分證相片規格驗證暨浮水印防偽系統」即產生,設計一套程式,提高換發國民身分證的工作效率及確保換證使用相片的正確度。並且延伸研究出使用內崁式的數位浮水印〈Digital Watermarks〉,將全國民眾的身分證照片統一建立資料庫,並自動加入個人資料浮水印。日後,照片只需透過本程式分析,即可知道其姓名、身分證字號、有無犯罪前科等個人資料。希望藉此達到降低偽造身分證之犯罪率,以保護民眾之權利。 The government is launching to renew national identification cards with new norms, to avoid fake ones. However, there are more than ten limits on photos, it could be wasting time to discern by people. Thus, I launched a research on "The xamination on new national ID card photos and watermark forgery-proof system". The program will help both to enhance efficient renewal process and to use correct photos. Also we developed the embeded Digital Watermark technology, which would create a database for ID cards of the nation and could add personal infomation automatically. With the help of the program, simply run the photo analysis, we could find out the names, ID number, criminal background, etc.We hope to decrease crimes via fake ID cards, and protect the national right.

聚苯胺導電高分子在鐵系金屬防蝕上的應用與研究

聚苯胺導電高分子的發展蓬勃迅速,其應用也愈趨廣泛,金屬防蝕為其可能應用之一。台灣四面環海,工業發達,造成金屬腐蝕損害嚴重。本文為探討聚苯胺導電高分子在鐵系金屬防蝕上之應用,針對以下各點進行研究: ( 1)探討導電高分子聚苯胺性質與合成方法(2)探討聚苯胺導電原理(3)測試不同腐蝕條件下聚苯胺的防蝕效果(4)探討聚苯胺防蝕原理(5)各種氧化還原態聚苯胺防蝕效果之比較。實驗結果顯示塗布聚苯胺與未塗布聚苯胺之鐵系金屬(生鐵、不鏽鋼、鍍鋅鐵)在 3 %氯化鈉、 0 . 5M 鹽酸、1M 鹽酸溶液中之腐蝕狀況,以鹼式中問氧化態聚苯胺( Emeraldine base 。)最具防蝕效果。聚苯胺防蝕機構包含阻隔防護、腐蝕抑制劑、陽極保護、惰性化、形成保護性氧化物薄膜、阻礙離子擴散速率、電化學介面遷移等機構 ·The development and application of the conducting polymer polyaniline is getting prosperous and popular. One of the applications is corrosion protection. Because Taiwan is surrounded by sea and the pollution of industry is more and more serious than before the damage of corrosion is greater and should be properly controlled. In this paper, the authors attempted to study the effect of the corrosion protection of polyaniline in ferrous metal. This study could be divided into four parts: (1) the study of the property and synthesis methods for the conducting polymer polyaniline, (2) the conduction theory of the polymer polyaniline, (3) The corrosion protection effects of the polymer polyaniline under different corrosion situations, (4) the corrosion protection theory of the polymer polyaniline, and (5) the comparison of the corrosion protection effects of various polymer polyanilines. According to the experimental results the Erneraldine-based polyaniline gets the best corrosion protection effect. The mechanism of corrosion protection may be the result of the following ones: barrier protection, corrosion inhibitor, anode protection, innobeling, chemical active layer. inhibition of diffusion rate, and the shift of electrochemical interface.

直接乙醇燃料電池之觸媒層研究

直接乙醇燃料電池以酒精與氧氣透過氧化還原反應產生電能,但化學反應緩慢,需利用觸媒以增加其速率。本實驗目的在於盡可能找出一個表現最佳的觸媒。本實驗利用活性碳粉作為觸媒(鉑、錫)的載體,以酸性(HNO3)與鹼性(NaOH)環境分別處理碳粉,再以含浸法與多元醇含浸法將觸媒還原。我們得到以HNO3 處理的碳粉無法保有原碳粉的型態,較利用NaOH 處理為差。在觸媒製備方面,多元醇含浸法還原效果比含浸法可得到較小的觸媒尺寸,在本實驗中,溶液中鉑與加入的碳粉重量比為3:7 時,可得到最大的反應面積。此外,當鉑與錫原子數比為4:1 時,可得到最大的乙醇氧化電流。Direct ethanol fuel cell is a kind of power source which generates electrical power by a redox reaction involving ethanol fuel and oxygen. However, this reaction takes place slowly; therefore, catalyst is needed to improve its activity. The goal of this project is to get an optimize catalysts ratio to obtain the best catalyst activity. Activated carbon is used as the support of catalyst (platinum and tin) particles in this project, which is pre-treated in acid (HNO3) and alkaline (NaOH) solutions respectively. Then, the precursor is reduced by impregnation and EG-impregnation. We learned that activated carbon pre-treated with NaOH activates better than which pre-treated with HNO3 because the latter bear less resemblance than the former. As for the catalyst, the results of EG-impregnation show smaller size of catalyst particles than those of impregnation. In this project, when the ratio of the weight of platinum and activated carbon added into the solution is 3:7, we can get the largest surface area. In addition, when the ratio of the amount of platinum and tin atoms is 4:1, we can get the largest current of ethanol oxidation.

豆類澱粉?抑制劑之研究與應用

豆類澱粉?抑制劑具害蟲防治與血糖調節之功效。本研究從台灣五種豆類中進行澱粉?抑制劑活性初篩,篩選出四季豆對麗蠅的澱粉?有明顯的抑制效果,粗萃後分離純化出單一蛋白質,經胺基酸定序結果得到兩個多?片段-VGLDFVLV 與TETSFNIDG , 與已發表文獻比對推測為腰豆澱粉?抑制劑—αAI-1。經測試發現此抑制劑在85℃時仍具備抑制果蠅澱粉?之活性,為一熱安定性蛋白質,其抑制作用受pH 值影響很大,在偏酸性環境下的效果最好,且其抑制作用具特異性,可明顯抑制果蠅、入侵紅火蟻、白蟻、蟑螂及麵包蟲等昆蟲的澱粉?活性。在調節血糖方面,本研究利用豬胰臟澱粉?進行抑制活性測試,篩選出四季豆、花豆與黑豆對豬胰臟澱粉?有明顯的抑制效果,進一步利用Wistar 品系大鼠,進行豆類澱粉?抑制劑降血糖功效之活體試驗,將四季豆、花豆和黑豆粗萃經濃縮乾燥後餵食大鼠,發現花豆、黑豆在第一小時具有顯著抑制血糖增加效果;四季豆、黑豆在第三小時血糖回升,花豆仍可維持較低血糖濃度。此初步結果顯示,花豆粗萃物可能具備減緩消化吸收作用,具應用潛能,其確切分子作用機制值得深入探究。Amylase inhibitors can be applied in pest control and glucose adjustments. The study screens Phaseolus vulgaris from five Taiwan’s beans that has the most significant inhibition towards Chrysomia megacephala. The protein was isolated and sequenced two fragments, -VGLDFVLV and TETSFNIDG, highly homologous to that of αAI-1 from Phaseolus vulgaris. The pure protein still inhibits the amylase from Drosophila melanogaster at 85℃, suggesting it is thermal-stable. Its activity was specific and was affected by pH, reaching the peak in weak acidic condition. It obviously inhibits amylases from D. melanogaster, Solenopsis invicta, Odontotermes formosanus, Periplaneta Americana Linnaeus, and Alphitobius sp. In addiction, the study points out Phaseolus vulgaris, Phaseolus coccineus L. and Glycine max, which have significant inhibition toward pancreatic amylase. The amylase inhibitors’ functions of decreasing in blood glucose were detected by animal experiments on Wistar rats. The rats fed with the extracts of Phaseolus coccineus L. and Glycine max showed the inhibition of glucose increasing at the first hour, while blood glucose concentration after rats fed with Phaseolus vulgaris and Glycine max increase at the third hour, and blood glucose after rats fed with Phaseolus coccineus L. maintains low. Its premier result indicates that the extract from Phaseolus coccineus L. might postpones digestion and has potential to be applied. According to these results, amylase inhibitors are worthy to further analysis.

聚苯胺導電高分子二次電池之研製及性能研究

塑膠一向被認為絕緣體。但Heeger,MacDiarmid,以及Shirakawa證實,塑膠可以被改製成電的良導體。這須在高分子合成的過程中,使碳鏈呈現單、雙鍵交錯排列情形。此外,高分子也須加以摻雜(doped),換言之,電子必須在氧化過程中被移除或在還原過程中被加進聚合物。電子移除所留下的電洞,或新加進的電子則可自由在分子鏈上移動,形成導電性。我們所要製備的導電高分子二次電池就是由這個原理完成的。所謂的二次電池(Secondary Cell)簡單的定義就是「可反覆充電、放電,循環使用的電池」,我們將會將其與市售電池比較,並改良出合適商品化的高分子二次電池。我們將以聚苯胺做為我們電池的正極,並用鋅片(原本我們要用鋰金屬的,但其再空氣中即易氧化,因此改採鋅)做為負極,製成一聚苯胺二次電池。The plastics is thought to be a insulator, but Heeger, MacDiarmid, and Shirakawa had proved that conductivity of plastics can be improved by doping other media. The conjugated polymers such as polyacetylene、polyaniline have the interlaced single-double bonds that electrons can move from one side to another side caused the production of electric currents. The purpose of this study is try to study the application of the conjugated polymer polyaniline to make a secondary battery. First, we use the chemical and electrochemical method to compose the polyaniline. Then we test the conductivity of the produced polyaniline and test the current and electrical potential of the polymer battery. The battery that we made from our laboratory has the electric potential about 0.5 volts and the electric current is greater than 50 microamperes. Finally, we also try to use the lithium as cathode to improve the performance of the polyaniline battery.

繪身繪影-正三角形磁磚設計方法與碎形密舖之研究

本研究主要以正三角形作為基本單元,透過窮舉討論得到正三角形邊的作用方式只有五種,再經由排列組合歸納出11 種正三角形密鋪磁磚設計方法。進一步,運用我們的研究結果,配合數學簡報系統製圖,創作新圖樣,也彌補了Escher 在手繪時所造成的誤差,達到完全密鋪的效果。碎形磁磚的部份,我們也依據其背後的數學理論創作幾套結構圖,利用結構解析,碎形密鋪磁磚將變得十分容易,學習者將可輕鬆製作富有創意的新圖樣。 ;This research mainly takes the regular triangle as the basic unit. Through the enumeration, we obtain that there are only five operations for edge of the regular triangle, and then 11 kinds of regular triangle design methods are induced. Even more, utilizing our findings and Mathematical Presentation System (Math PS), we created the new pattern which makes up Escher’s errors and achieves the tiling. As to Fractal Tiling, we create several sets of structure drawings according to its mathematics theory. Using structure analysis, the Fractal Tiling will become extremely simple, and the learner can make the rich creative new pattern easily.
