趨光性及族群壓力的探討

The purpose of the research was to find the relationship between the fruit fly’s attraction to light (or heliotaxis) and their genes. Fruit flies, because of their short life span of approximately two weeks, and their simple requirements for survival, are ideal candidates for the study. With a specially designed device "Mi Gong" for the experiments, fruit flies could be separated and cultivated in accordance to their preference to specific wavelengths of light. They received cross-breeding and were studied to learn the ratio of the resulting offspring in their preference to the light. It was observed that the majority of the fruit flies were more attracted to violet light. It was also found that the next generation of the fruit fly inherited higher sensitivity to the light than their parents. Knowing that there could be potential margin of errors in the experiments, the results of the study demonstrated that light, as an external source, had limited impact to heliotaxis of fruit fly. The study suggested that insects with heliotaxis, including fruit fly, inherited heliotaxis in their genes. The study also pointed out the potential benefits of employing heliotaxis in many areas especially in the agricultural development. Furthermore, heliotaxis can be utilized to study the difference in the behavior of nightlife insects before and after the invention of manmade light source.這項研究,主要是在探討果蠅的趨光性和遺傳的關係。很多種昆蟲都具備趨光性,我們之所以以果蠅作實驗,主要是因為其生活週期短,大約兩週,且易於培養。實驗內容是以「迷宮器材」來鑑別果蠅對某一特殊色光的偏好,將該群果蠅分離出來培養,令其繁衍後代,觀察其後代對此一色光偏好的比率,我們所用的果蠅大致上對綠色光較敏感,而其子代的敏感度較親代高。因此,我們推論趨光性會遺傳,且受光影響,但並非全由光影響,故這可能是實驗中的誤差成因。雖然此實驗是以果蠅為對象,但亦可假設其它具趨光性的昆蟲,也可能是因遺傳而導致其子代有趨光性,而我們可利用其趨光的一些特性,減少農作物因害蟲類的果蠅而損失,並且希望可以得知,在人造光源還未出現在世上時,夜行性昆蟲是否有差異。

應用垂直循環層流淨化水質之研究

The results of the thermal diffusion experimental show that the separation efficient\r increases with increasing ΔT, but decreases with increasing Q at C0 constant. When\r Q is kept constant, the efficient of change ΔT are small, and the maximum at C0 about\r 545 mg/L. The purification efficient of raw water for thermal diffusion is low.\r And it is high energy consumption, difficult operation, and high costs. So the potential\r of the application is low. Therefore, the vertical circulatory laminar flow apparatus\r was developed in this study. And the separation equation of this apparatus was also\r set up. The results of vertical circulatory laminar flow experimental show that\r the efficient increase with increasing V, and the optimum conditions are at about\r Q= 1.05 L/min, C0=800±50 mg/L, and V=26 cm/min. The separation efficient of the\r vertical circulatory laminar flow apparatus is higher than thermal diffusion apparatus.\r And its energy consumption and costs are low, and easy to operate. Since all interior\r moveable plates have two operation sides, so the multiple channel apparatus is application.\r Therefore, the multiple channel apparatus can treat large amount raw water and has\r low costs. The efficient of the experimental all are better than the theoretical\r values that may due to: (1)the assumed partical diameter is too small,(2)the flow\r is not real laminar flow, and (3)the end-side effect is not considerate. \r 熱擴散實驗結果顯示在固定C0下,分離成效隨ΔT增加而增高,但隨進料流率增加而降低。在固定ΔT下,分離效率亦隨進料流率增加而降低。固定進料流率下,增加ΔT對分離效果的影響不大,以C0=545±10\r mg/L時,顯現出最佳成效。採用熱擴散裝置雖對降低原水中之懸浮微粒含量效果有限,且能源需求高,造成高成本和操作複雜,不具實用潛力。為提高分離原水微粒之成率,以及簡化操作與降低成本,本研究開發垂直旋轉帶裝置來製造出上下循環水流,並導出水流與微粒於其中之流速分布式,以及頂部、底部出口之微粒濃度計算式。垂直循環轉帶實驗結果,顯示提高V值有助於降低Ct/Cb比值;Q=\r 1.05 L/min左右、C0=800±50 mg/L下與V在26 cm/min左右,均有較低之Ct/Cb比值。垂直旋轉帶裝置分離成效明顯優於熱擴散裝置,且設備簡單、能源需求低與操作容易。由於旋轉帶之兩面均可以做為移動平板,因此不僅可以用來處理大量之高濁度原水,也能更具經濟效益。由理論與實驗結果之比較,顯示理論成效較實驗差,探討其原因可能是:(1)平均粒徑之假設值偏低、(2)流場並非全然以層流流動、(3)兩端終端效應未加考慮等。

“氮”憑本事-土壤中單棲固氮細菌族群比例及親緣關係探討

Azotobacteraceae 為一單棲固氮菌科,包含Azotobacter 與Azomonas 兩菌屬,在農業上可用來改善缺氮的貧瘠土壤。在分離土壤中的Azotobacteraceae 時,發現非單棲固氮菌與單棲固氮菌間可能具有共生的情形。我們利用優勢培養(缺氮)的方法篩選土壤中的Azotobacteraceae,將優勢培養後所生成的菌落稀釋104~106倍後,能有效分離Azotobacter 與Azomonas,然而低於此稀釋倍率則會形成混合菌落,其中可同時發現單棲固氮菌與非單棲固氮菌存在,推測某些非固氮菌在優勢培養過程中可能可從單棲固氮菌獲得氮源,與之共生。此外亦從菌種形態的差異並配合顯微螢光雜合技術(fluorescence in situ hybridization, FISH)、分子遺傳標記(16S-rDNA)等方式,分析土壤中的Azotobacteraceae,探討單棲固氮菌及其他非單棲固氮菌在培養基上的生長情形、比例及親緣關係。The family Azotobacteraceace is group of free-living nitrogen-fixing bacteria that is found in soil. Two genera are within this family: Azotobacter and Azomonas. Agriculturally, it is often used to improve fertility for nitrogen deficient barren lands. We analyze the Azotobacteraceace according to molecular biology and traditional taxonomy. We used an enrichment procedure to culture the bacteria, and diluted it repeatedly. We found it most suitable to dilute it 104~106 times to best separate Azotobacter from Azomonas. If the concentration were to be higher than this, mixed flora containing many different bacteria species would be found. Moreover, we noticed that non nitrogen-fixing bacteria, symbiotic nitrogen-fixing bacteria, and free-living nitrogen-fixing bacteria would form a single colony on a nitrogen-deprived medium. This implies that a symbiotic relationship may exist between nitrogen-fixing bacteria and non nitrogen-fixing bacteria. We also discuss the growing situation, the group proportion, and the relationships between free-living nixtron fixing bacteria and other bacteria by morphology, fluorescence in situ hybridization (FISH), and molecular biology.

日長處理對大豆蛋白質合成的影響

日照影響花芽分化及生長試植物生理學上的重要課題,根據相關文獻指出,蛋白質類型的變化是影響開化的重要關鍵,大豆是短日照植物,分別以長日照(LD18hr/DAY)和短日照(SD6hr/DAY)處理,抽取葉片中的蛋白質以SDS-PAGE.2D-SDS-PAGE分析,SDS-PAGE的實驗姐果顯示出在分子量25.2 32.9 38.6 20.6 64.0 100.6 kDa處中的蛋白質SD比LD的量多,但是在分子量16.8kDa以下的蛋白質LD含有高濃度而自SD只有一點,我們使用2D-SDS-PAGE做進一步的分析,SD與LD分為三個區塊討論(SD1-3 LD1-3)有許多蛋白自LD1.LD3出現,但在SD1 SD3中未出現,可能有抑制花芽分化的作用,而在SD2中有多個高濃度蛋白出現,可能在花芽分化上扮演重角色,其中編號P1 P2的蛋白質有較明顯的變化,根據NCBI網站上的資料顯示,P1蛋白質與光敏素C有關,而P2蛋白質則沒有找到分子量等電點類似的蛋白質;The effects of photoeriod on floral initation and develoment is an imporant phyiological subject and have studed ve he past 70 years. Accrding to the reference, polypeptide partten plays an important rolein photoperiodic induction. Soybean s a SD(short day)plant. So we planted them with 6-h(short day,SD)and 18-h(long day,LD)photoperiods. Total proteins n leaves of soyban were extracted from the vegetable,SD and LD. And using SDS-PAGE and 2-D eloctrophoresis to analyze. 25.2,32.9,38.6,50.6,64.0,and 100.6kDa polypeptides in SDS-PAGE were always present in leavess at SD but only a little bands f the polypeptides appeared at LD. And many polypeptides whose molecular weight(Mw)is below 16.8 kDa had higher concentration at LD ad Reduced at SD. We found three different areas(SD1~3,LD1~3) and analyze them. Many proteins were found n LD1,LD3 and reduced in SD2,SD5. Possibly, these polypeptides found in LD1 and LD3 play an important role in restraining floral iniiation in soybean. Many proeins had more concentration in SD2. Probably, they play an important role in photoperiodic induction and floral initiation. We found a protein labeled P1 in SD2 and it is with molecular weight(Mw) of 36kDa at isoelectric point(pI) of5.2, and it may be related to cytochromeC. We also found protein labeled P2 in SD2 and it is with molecular weight(Mw) of 40.2kDa at isoelectric point(pI)of 5.4, and we couldn't find any protein that has the similary Mw and pI in potein database about flowering and daylength.

核醣核酸蛋白粒之K 蛋白基因表現及功能之研究

核糖核酸蛋白粒之K 蛋白具有多種功能,可參與在基因轉錄和蛋白質轉譯等過程。我們發現\r 一新奇的cDNA 較已報告的K 蛋白cDNA 少了72 個核?酸,分別命名為S 形與L 形。由基因\r 組DNA 的分析,S 形可能是經由替代剪接少了第8 個exon 所形成。以西方墨點法分析細胞中\r 的K 蛋白,亦證明有S 形存在,其表現量較少且多存在於細胞核中。為了探究兩種K 蛋白等\r 形的表現和功能,我們以PCR 偵測K 蛋白在不同組織與細胞株和人類乳腺腫瘤組織中的RNA\r 表現,結果顯示主要以L 形為主;我們並將S 形與L 形K 蛋白分別轉染至人類乳癌細胞株MCF7\r 中,以抗生素篩選出穩定表現K 蛋白之細胞株,生長速率分析顯示表現S 形之細胞株生長速\r 率較慢,其分子機轉仍有待研究。另外在比對K 蛋白之基因組DNA 時,除了在第九對色體\r 上有完整之K 蛋白基因組DNA,我們亦發現在第2、3、5 及11 對染色體上有類似K 蛋白基\r 因的序列,以cDNA 的形式存在,這些DNA 序列是否可以表現RNA 及其意義為何,目前尚\r 不清楚。Heterogeneous nuclear ribonucleoprotein K (hnRNP K) is a multifunctional protein known to be\r involved in the regulation of transcription, translation, nuclear transport and signal transduction. We\r have identified an alternative splicing transcript of hnRNP K lacking exon 8. This novel isoform\r (named S form compares to L form of full-length hnRNP K) encodes a protein that is 24 amino acids\r depletion between RNA binding KH1 and KH2 domain. To explore the functional roles of both isoforms,\r we detect their expression in several tissues (including liver, lung, kidney and heart), cell lines (MCF7,\r 293T, HeLa, NIH3T3, WEHI, P388D1) and specimens of human mammary gland tumor by RT- PCR.\r The results showed that L form was expressed in all samples, whereas S form was only expressed in cell\r lines. Using Western blotting analysis, we found that L form existed in both cytosol and nuclear\r fractions, and little amount of S form was detected in the nuclear extract. Furthermore, we construct the\r MCF7 cell lines that stably expressed S form or L form hnRNP K. Growth rate analysis indicates that\r the overexpression of S form of hnRNP K could decrease cell growth rate. The molecular mechanisms\r of growth inhibition by S form hnRNP K are to be further investigated. On the other hand, when we\r blast the human genomic genebank, we found except the chromosome 9 containing complete hnRNP K\r genomic DNA, there are near complete hnRNP K cDNA sequence appears in chromosome 2, 3, 5, and 11.\r The meaning of these sequences is unclear.

環境因子影響美洲蜚蠊觸角擺動模式之研究

本研究以攝影紀錄的方式,透過電腦進行影像分析,記錄不同刺激下美洲蜚蠊(Periplaneta americana)的觸角擺動模式,計算出各項觸角運動的參數,以瞭解光線(光刺激或光適應)、震動刺激、喝水與進食對其觸角行為的影響。我們發現在不同因子的刺激下,觸角擺動的模式具有差異,若兩種不同的刺激同時發生,蜚蠊觸角的行為亦具整合性的反應。蜚蠊於不同狀態下(如喝水或進食),對相同的刺激有不同的反應,證明蜚蠊觸角的行為模式,受環境因子與個體狀態調節。透過掃瞄式電子顯微鏡的觀察,也發現觸角具多種感覺毛,且雌雄的感覺毛的分佈與數量具有差異。綜合以上發現,證明觸角除了為敏感的受器,亦為能反映出生理與環境狀態的動器,同時也適合進行發展檢測器的仿生學應用,用來檢測環境中物理及化學因子。The aim of this study is to investigate the different swing motion modes of antennae of American cockroach (Periplaneta americana) by computer-aided Imaging Analysis. The parameters of each swing movement were calculated in order to analyze how light (including light stimulation or light adaptation), vibration, food and drinking water may affect the antennae behavior of American cockroach. It was found that the antennae swing motion modes were significantly different under different types of stimulus. If two different types of stimulus occurred at the same time, the reactions of antennae motion may become conformable. Under different environmental conditions (such as food or water), same stimulus may result in different reactions. The antennae behavior has shown to be significantly affected by environmental conditions and individual physiological status. Through the observation with scanning electron microscope (SEM), it was found that the antennae has many types of sensilla; and the distribution and quantity of these sensilla are significant different between sexes. In conclusion, not only the antennae are considered as the sensitive receptors, but also they are the important effectors to reflect physiological status and environmental conditions. The current model is suitable for the development of specific detectors in the applications of Bionics to detect the physical and chemical factors in certain environments.

外觀數列

The Look and Say sequence is produced by describing the appearance of the previous row. For example, start with “1,” which can be described as “one 1,” and therefore the second row is “11,” which is "two 1s," making the third row “21,” the fourth row “1211,”and so on. The main goal of this study is to work out the exact formula for this sequence, which means given the row number n, we can know at once what the n-th row is without having to start from the first row and doing the look-and-say iteration for n-1 times. Some of the methods used include dividing groups, repetition and cracks. The formula we derived speeds up the calculation and gives us a better understanding of the look and say sequence.「外觀數列」為依照外觀產生下一列的數列,第一列為「1」,第二列描述第一列「1 個1」而為「11」,第三列則描述第二列「2 個1」而為「21」,第四列「1211」,依此類推。本研究針對外觀數列的各項數學性質作研究探討,並由此推導出外觀數列的一般式,即給定第n 列就可知道該列的內容。我們運用了分組、重複性以及裂縫的方法分析數列,最後得到了其一般式,此一般式有助於運算速度的加快以及我們對數列性質的了解。

Transformer Robot

Nicholas’s project is a transforming robot, who he has named Reggie. He's about a foot tall and can transform from a biped into a vehicle and vice versa?\r Reggie has been Nick’s project for just under a year now, mainly working on him in technology classes. He operates without physical connection to anything else, so everything he needs from micro controllers to power is stored somewhere on Reggie’s body?\r Reggie is controlled through a programmable Arduino board (programmed in C++) that is connected up to a motor driver, for controlling the two driving motors and an SSC-32 servo controller for controlling the 14 servos via serial port. The Arduino uses the SSC-32 to coordinate the servos movements using pre-programmed sets of movements stored in procedures in order to complete all actions.\r All the parts were produced on a 3D CAD system to begin with, as well as the robot as a whole, although the design did change drastically over the course of its construction and now is considerably different from the original design as there were many problems and complications ranging from space to torque issues that called for a change in design.\r Most of the parts were from the servo erector set from lynx motion, although some of them were used in the way they were intended to and most of them were modified slightly. Some parts Nick manufactured himself either on a rapid prototyping machine, a lathe or milling machine and sometimes a combination of them or even high temperature soldiering?\r Currently Reggie is capable of driving and standing up and is currently radio controlled. He will eventually be capable of walking and modifications to allow him to (such as stronger leg servos) have been made to facilitate that, but it is possible more may have to be made.

Nonlinear Time Series Analysis of Electroencephalogram Tracings of Children with Autism

Methods of nonlinear time series analysis were compiled for use in the analysis of Electroencephalogram (EEG) tracings of children aged three to seven with varying degrees of autism in order to provide a quantitative means of diagnosing autism and determining its severity in a child. After determining the EEG leads to be used for analysis, the identified methods were coded and saved as functions on Scilab. To test the compiled program, a minimum of five EEG readings per cluster of children diagnosed with mild, moderate, severe and no autism will be obtained. The project was able to identify the mean, standard deviation, skewness, kurtosis and other higher order moments, the autocorrelation function, and the Fourier Series as the time-resolved statistical methods to be used for time series analysis. The nonlinear analysis methods identified include the use of the correlation integral, time-delay embedding and the Lorenz equations. One-way ANOVA testing will then be used on the numerical data obtained from the analysis to determine if a significant numerical differentiation has been obtained between the different clusters of EEG. This will provide a definitive way to medically diagnose autism, pinpointing children afflicted with the disorder and giving them proper treatment.\r Two copies of the "Abstract of Exhibit" (in English) should be sent to the National Taiwan Science Education Center or email to fung@mail.ntsec.gov.tw or yuonne@mail.ntsec.gov.tw before December 31, 2009.

神經妙算流博村-是洪水?是山崩?

南投溪頭米堤飯店在2001 年因土石流受創,產險公司以「土石流即是山崩」為由拒絕理賠。歷經5 年的訴訟後,2007 年1 月台灣高等法院認定,土石流屬保險契約中所規範的「洪水」,並非「山崩」,判決產險公司應給付賠償金。土石流究竟是洪水?是山崩?主要因素便是土石流發生的水文因子。本文藉由博愛村的現場調查及文獻探討先作初步資料分析(preliminary analysis),了解到地文因子(physiographical factor)是土石流發生的充分條件,非必要條件;一般僅考慮雨量因子,把地文因子看作常數(忽略地文因子受水文因子歷程(course)影響),簡單易懂,但在安全與經濟考量上有待討論。使用類神經網路對已發生過的土石流事件計算土石流發生臨界曲線,並使用模糊理論計算松鶴地區其受水文因子影響的土石流發生臨界曲線,這樣的模式,考慮似乎比較周到。由「米堤飯店」的例證,更說明土石流發生的水文因子的必要性。Lemidi hotel, Xitou, Nantou because mudflows and landslides and was wounded in 2001. The Insurance Company refused to settle a claim on the position on “A landside is a debris flow”. After the lawsuit which was going through 5 years. In January, 2007, the High Court of Taiwan asserted that the adversity in the 2001 belongs to “the flood” in the norm of the insurance agreement. As the norm of the insurance agreement said, the debris flows in the land is the flood, not the “landslide”. The High Court of Taiwan judged that the Insurance Company should compensate the Lemidi Hotel. Is debris low a landslide or a flood? The main cause is the Hydrology in the happening of a debris flow. We did preliminary analysis of forms by on-the-spot investigation on Song-Ho Village and reference discussing. We realized that physiographical factor is an abundant condition for a happening of a debris flow, not the essential condition. Generally, people only consider rainfall factor. And they consider physiographical factor as not as constant (neglect the influence of hydrology factor to physiographical factor). We can understand easily in that way. Therefore, in the aspect of security and economy, there is much doubt that is needed to be discussed. People who use neural network method to calculate the curve of the debris flow happening , and used fuzzy theory to calculate the curve of the debris flow happening which is influenced by the hydrology in Song-Ho Village. In that way, we may consider more thoughtful. From the example of the Lemidi Hotel in Nantou, we can prove that how necessary the hydrology factor in the debris flow is.

超音波對星狀細胞產生神經營養因子的影響

超音波應用於臨床已行之有年,而以穿透式超音波打入腦部的想法也已提出,本實驗目的即為測試低強度間歇性超音波達到腦部受傷後治療的可能。本實驗採用星狀細胞培養模式來探討,首先以腫瘤壞死因子α(Tumor Necrosis Factor-α, TNF-α)模擬中風缺氧的狀態,並利用低強度間歇性超音波刺激星狀細胞,然後檢測其培養液中一氧化氮(nitric oxide, NO)的濃度變化,以及大腦衍生神經營養因子(Brain-derived Neurotrophic Factor, BDNF)、神經膠質衍生神經營養因子(Glial-derived Neurotrophic Factor, GDNF)之mRNA 表現的情形。由實驗結果顯示,在TNF-α存在狀況下則能使NO 濃度上升,施打低強度間歇性超音波可促使NO 增加,並提升上述BDNF、GDNF 兩種蛋白質之mRNA 表現,BDNF 可幫助支持神經元生存,促使新神經元和突觸的發展及分化;GDNF 可有效提昇許多類型神經元生存率;而適當之NO 濃度可以調節神經傳遞物質釋放和腦血流,乃至於影響到記憶的產生。我們初步的結果也顯示在TNF-α存在下,超音波可以增加BDNF 及GDNF 蛋白之產生,因此本研究成果顯示低強度間歇性超音波可提供有利於神經元的環境。The ultrasound technology has been widely applied in the clinical practice for many years, such as monitoring of fetus, lithotripsy, rehabilitation after fracture, as well as the ultrasound stimulation of brain tissue, etc. The purpose of this study is to investigate the therapeutic effect of low intensity pulsed ultrasound (LIPUS) on the injured brain. In this study, we adopted the in vitro model of astrocyte cultures. The tumor necrosis factor- α (TNF-α) was used to simulate the condition of ischemic stroke. We used LIPUS to stimulate the cultured astrocytes, then we ssessed the concentration of nitric oride (NO) in the culture medium. We also assessed the expression of messenger RNA (mRNA) of both brain-derived neurotrophic factor (BDNF) and glial cell-derived neurotrophic factor (GDNF). The results showed that NO increased after the addition of TNF- α. In addition, treatment of LIPUS increased the expression of NO, as well as the mRNA of both BDNF and GDNF. The BDNF is able to support the survival of neuron, as well as stimulate differentiation of neurons and synapses. GDNF can also increase the survival of various kinds of neurons. Furthermore, NO may regulate the secretion of neurotransmitters, increase the blood flow in the brain, as well as enhance the processing of memory. Our preliminary results also demonstrated that both BDNF and GDNF protein increased in response to LIPUS in the presence of TNF-α. Therefore, treatment with LIPUS implicated an environment favoring the protection of neurons.

CryptoDefender - afast way to encrypt your files

1. Purpose of the research\r Nowadays, many encryption software are available to computer user. However, very few people use them which result in many cases of information and data leakage due mainly over the Internet. Most people use USB Flash Drive to store and carry confident files back home to work on. When the USB Drive is lost, the files stored are often leaked. To prevent this type of incident occurring, “CryptoDefender” is developed to enhance files protection and to change user’s habit in handling confidential information. \r 2. Procedures\r “CryptoDefender” is an add-in for common application software, e.g. Word, Excel, etc. It will appear as a new tab in the ribbon which provides the software an alternative for saving and opening a file. It protects the files in two ways: encryption and steganography. The data is first encrypted into cipher text as these encrypted data will be hidden in an image file to lower suspicion of readers. All of these procedures will be done by one click. For encrypt and decrypt, the user just needs to click the “Protect” and “Decrypt”. An extremely fast way to encrypt the file is “Quick Protect”. It will randomly choose an image from the image folder according to their file size. This function saves the time for choosing an image.\r 3. Data\r The method for steganography is using last two bit of R, last three bit of G and B to store the encrypted data. In 24-bit bitmap, RGB is represented the color of each pixel and each of them is 8-bit. The leftmost bit of each byte contains the highest value, and the right one contains the lowest value. Therefore, any change in the rightmost byte will only cause a minimal change in a byte value, and the color as well. 2-3-3 method is adopted to replace the pixels as encrypted data. It is used because it is in coordination with the program’s fluency. 2-3-3 is obtained so that 1 pixel will contain 1 byte of encrypted data.
